However I want to know how performance(opening duration) will be affected shared office files (up to 250 MB).
Note: 1Gbps network access is available at the client PCs.

I guess you are asking about cache or qtier (QNAP NAS model and firmware were sadly not disclosed)
Only files that would be read over and over and over again would benefit from a cached state. As they still would need to be read from disk if they are accessed only once.
On top office files in a 1GbE environment would also not benefit from write cache
So, save the money and the hassle
TS-H1277AXU-RP is the NAS model.
Actually, read performance improvement will be sufficient.
I also want to learn that whether file caching relies on the file name or not.
Case 1 : user accessed a file, changed it and saved it .
Case 2 : user accessed a file, changed it and saved it under a new name.
Will that file be stored in the cache ,and can we benefit from read cache mechanism on the 2nd access at both of these cases too,

Cache is block based, so whatever blocks are accessed ‘often’ would land in the cache.
Either try it out or just read up on ZFS cache mechanisms, again for office files probably a waste of time and effort.
